The work of our team at Roche has been published in a paper on selective Aldosterone Synthase compounds and even made it on the title picture.. Our multiplex-LC/MS assay for simultaneous quantifications of six steroid intermediates and hormones allowed the determination of CYP11B1 – B2 selectivity in-vitro and in-vivo, being instrumental for the development of an over 100fold selective CYP11B2 inhibitor, that suppressed effectively aldosterone production without impairing the cortisol response upon ACTH challenge.
This is my most beautiful example of how a translational target response biomarker measurements can bring forward difficult projects, and of how useful LC/MS can be in discovery biology.
